      Detection of mouse cleaved Bid by immunoblotting. Samples: Whole cell lysate from L929 cells left untreated (lane 1), treated with actinomycin alone (5 µg/ml, lane 2), or treated with 5 µg/ml actinomycin plus 2.5 ng/ml, 5 ng/ml, and 10 ng/ml mouse TNFα (lanes 3, 4, 5, respectively).       DescriptionImmunoaffinity purified rabbit polyclonal antibody. Recognizes the ~15 kDa cleaved mouse Bid.
      BackgroundBid (~22 kDa) is a BH3 domain containing pro-apoptotic cytosolic protein that maps to human chromosome 22q11.2. Initially localized to the cytoplasm, the Bid protein is cleaved by the action of caspase 8 into fragments of two different sizes, p15 and p13 and becomes inserted into the mitochondrial membrane. Of the two fragments the p15 BH3 containing fragment is the more potent inducer of apoptosis and in fact is a stronger inducer than the full-length Bid protein. Following cleavage by caspase 8, Bid will bind with Bax localizing it to the mitochondrial membrane and leading to the release of cyto-chrome c. This interaction is blocked by Bcl-x, which directly binds to Bax, competing for the binding of Bax to the membrane-localized Bid.
