      Application CommentsFor an MMP-1 or MMP-3 positive control, incubate HT-1080 cells for 2 hours at 37°C in serum-free media containing 100 nM PMA. Collect and centrifuge medium; concentrate as necessary. Store in aliquots at -20°C. When staining paraffin sections for MMP-1, pretreatment with heat, saponin, or trypsin can be used, but a pressure cooker generally gives the best results. Anti-MMP-9 (Ab-1) recognizes both the latent and active forms of MMP-9 under non-reducing conditions but only the latent form under reducing conditions. Anti-MMP-9 (Ab-1) will inhibit MMP-9 enzymatic activity. Anti-MMP-9 (Ab-2) recognizes only the latent form of MMP-9 (non-reducing or reducing conditions). Anti-TIMP-1 (Ab-1) will react with TIMP-1 and MMP/TIMP-1 complexes. Antibodies should be titrated for optimal results in individual systems.

      BackgroundMatrix metalloproteinases (MMPs) are a family of enzymes that are responsible for the degradation of extracellular matrix components such as collagen, laminin and proteoglycans. All MMPs share: sequence homology, a catalytic mechanism dependent upon a zinc ion at the active center, ability to cleave one or more extracellular matrix components, secretion as zymogens which are activated by removal of an approximately 10 kDa segment from the N-terminus, and regulation by endogenous inhibitors. These enzymes are involved in normal physiological processes as well as in metastasis. MMP-2 and MMP-9 are members of the gelatinase family and elevated levels may be correlated with metastatic potential. MMP-1 is also known as interstitial collagenase and MMP-3 as stromelysin-1. Activation and activity of the MMP's are regulated by the TIMP (tissue inhibitors of metalloproteinase) family members. TIMP-1 preferentially regulates MMP-9 and TIMP-2 regulates MMP-2.
