Background InformationTks5 (previously known as Fish), is a large scaffolding protein with one amino-terminal PX domain and five SH3 domains. It is cytoplasmic in normal fibroblasts. In Src-transformed cells, Tks5 localizes to podosomes. The coexpression of Tks5 and constitutively active Src in epithelial cancer cells resulted in the appearance of podosomes/invadopodia. Cells with reduced Tks5 expression are poorly invasive through matrigel. Tks5 is expressed and localized to podosomes/invadopodia in some invasive human cancer cell lines and tumor tissue, particularly breast cancers and melanomas. Thus, Tks5 appears to be required for podosome/invadopodia formation and for invasion of some cancer cells.

      UniProt Number
UniProt SummaryFUNCTION:Required for podosome formation, degradation of the extracellular matrix, and for the invasiveness of some cancer cells. Binds phosphatidylinositol 3-phosphate (PtdIns(3)P) and phosphatidylinositol 3,4-biphosphate (PtdIns(3,4)P2). In association with ADAM12, mediates the neurotoxic effect of beta-amyloid peptide By similarity.

SUBUNIT: Interacts with ADAM12, ADAM15 and ADAM19 By similarity.

LOCATION: Cytoplasm. Cell projection › podosomeBy similarity. Note= Cytoplasmic in normal cells and localizes to podosomes in Src-transformed cells By similarity.

Tissue specificity Widely expressed. Not found in the spleen and testis. Ref.1

Domain The PX domain is required for podosome localization, and for binding phosphatidylinositol 3-phosphate (PtdIns(3)P) and phosphatidylinositol 3,4-biphosphate (PtdIns(3,4)P2) By similarity.

The fifth SH3 domain mediates binding with ADAM12, ADAM15 and ADAM19 By similarity.

PTM: Tyrosine phosphorylated by SRC. Phosphorylation plays a regulatory role in the protein localization. The intramolecular interaction of the PX domain with the third SH3 domain maintains the protein in the cytoplasm and phosphorylation disrupts this interaction, resulting in the redistribution of the protein from cytoplasm to the perimembrane region. Phosphorylated on serine upon DNA damage, probably by ATM or ATR By similarity.

SIMILARITY: Belongs to the SH3PXD2 family.

Contains 1 PX (phox homology) domain.

      Contains 5 SH3 domains.

Molecular Weight130-150 kDa. Uncharacterized bands(s) may be observed in some cell lysates.
