      Applications
ApplicationAnti-Metabotropic Glutamate Receptor 2/3 Antibody detects level of Metabotropic Glutamate Receptor 2/3 & has been published & validated for use in IH(P), IH & WB.
      Key Applications
      • Immunohistochemistry (Paraffin)
      • Immunohistochemistry
      • Western Blotting
Application NotesWestern blot analysis can be done at final concentrations of 0.1-0.5 μg/mL. AB1553 can be used for immunocytochemistry using paraformaldehyde or paraformaldehyde/glutaraldehyde fixed tissue with light and electron microscopy. Cryostat and vibratome sections can be used with or without Triton X-100 treatment. Final concentrations recommended are 0.1-0.5 μg/mL.

The antibody has not been tested for use in immunoprecipitation. However, antibodies to the carboxy termini of other glutamate receptors are very effective in immunoprecipitation.

Optimal working dilutions must be determined by end user.
      Biological Information
ImmunogenCarboxy terminus peptide of rat mGluR2 conjugated to BSA with glutaraldehyde (NGREVVDSTTSSL).
      HostRabbit
SpecificityMetabotropic glutamate receptors mGluR2 and mGluR3, which have nearly identical C-termini. There is also a very slight recognition of mGluR5. Western blot analysis of rat brain shows bands migrating at Mr = 100,000 (which contains both mGluR2 and mGluR3) and 190,000. The larger band is thought to be a dimer of the smaller band.

      Entrez Gene Number
Entrez Gene SummaryL-glutamate is the major excitatory neurotransmitter in the central nervous system and activates both ionotropic and metabotropic glutamate receptors. Glutamatergic neurotransmission is involved in most aspects of normal brain function and can be perturbed in many neuropathologic conditions. The metabotropic glutamate receptors are a family of G protein-coupled receptors, that have been divided into 3 groups on the basis of sequence homology, putative signal transduction mechanisms, and pharmacologic properties. Group I includes GRM1 and GRM5 and these receptors have been shown to activate phospholipase C. Group II includes GRM2 and GRM3 while Group III includes GRM4, GRM6, GRM7 and GRM8. Group II and III receptors are linked to the inhibition of the cyclic AMP cascade but differ in their agonist selectivities.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q14416 # Receptor for glutamate. The activity of this receptor is mediated by a G-protein that inhibits adenylate cyclase activity. May mediate suppression of neurotransmission or may be involved in synaptogenesis or synaptic stabilization.
SIZE: 872 amino acids; 95568 Da
SUBUNIT: Interacts with GRASP (By similarity).
SUBCELLULAR LOCATION: Cell membrane; Multi-pass membrane protein.
TISSUE SPECIFICITY: Widely expressed in different regions of the adult brain as well as in fetal brain.
SIMILARITY: SwissProt: Q14416 ## Belongs to the G-protein coupled receptor 3 family.
