      Background InformationPotassium channel, subfamily K, member 16 (UniProt: G5E845; also known as KCNK16, TALK1) is encoded by the Kcnk16 gene (Gene ID: 74571) in murine species. KCNK16 is a multi-pass membrane protein that contains two pore-forming P domains. It is an open rectifier that primarily passes outward current under physiological K+ concentrations. KCNK16 is predominantly expressed in the pancreas and is activated at alkaline pH. Mice deficient in KCNK16 display reduced food intake and elevated oxygen consumption and energy expenditure. Islets with Kcnk -/- show enhanced insulin secretion and GIP-1 stimulation. KCNK16 is also reported to reduce delta-cell cytosolic Ca2+ elevations and somatostatin release by limiting their Ca2+-induced Ca2+ release and can modulate the intra islet paracrine signaling mechanisms that control glucagon secretion. KCNK16 is also reported to regulate beta-cell endoplasmic reticulum (ER) Ca2+ handling by conducting K+ counter currents across the ER membrane that promotes ER Ca2+ leak.
