      Background InformationMillipore’s Ready-to-Assay GPCR frozen cells are designed for simple, rapid calcium assays with no requirement for intensive cell culturing. Millipore has optimized the freezing conditions to provide cells with high viability and functionality post-thaw. The user simply thaws the cells, resuspends them in media, and allows them to recover for 24 hours. The following day the cells are counted, plated, and assayed for calcium response.

      The histamine H3 receptor was defined pharmacologically to be a presynaptic histamine receptor that binds selectively to R-(α)-methylhistamine and inhibits neurotransmitter release in the central and peripheral nervous systems (Hill et al., 1997). Molecular cloning of the H3 receptor confirmed it to be a Gi-coupled receptor widely expressed at high levels in the brain (Lovenberg et al., 1999). H3 presents a potential therapeutic target for disorders of the central nervous system involving attention, cognition and feeding (Leurs et al., 2005). Millipore’s cloned human H3-expressing cell line is made in the Chem-3 host, which supports high levels of recombinant H3 expression on the cell surface for functional detection via the calcium signaling pathway. Thus, the cell line is an ideal tool for screening for agonists, antagonists and modulators at H3.
