      Application NotesWestern blot: 1 μg/mL of the antibody will detect p73 in 30μg of total cell lysate from overexpressing cell lines. Higher levels of the antibody may be necessary when using native tissue sources.

      Immunoprecipitation: Add 1μg of antibody to a sample that has been precleared with Protein A Sepharose™ in 500 μl lysis buffer. Incubate antibody with sample at 4°C for 2 hours with agitation. Follow by incubation with 50 μL Protein A Sepharose™ for 2 hours at 4°C with agitation. Collect the Protein A complex by centrifugation, washing pellet 3 times in lysis buffer. Resuspend final sample in electrophesis buffer, denature by heating at 95°C for 5 minutes, and analyze by SDS-PAGE

      Immunohistochemistry of paraffin sections: Signal amplification may be necessary, in conjunction with standard IHC procedures. Dewax and rehydrate sections through descending graded alcohols to water using standard dewaxing protocols. Subject section to heat antigen retrieval. Wash sections 3 times with TBS, pH 7.4. Block endogenous peroxidase activity by incubating the section in 1% H2O2 in methanol for 30 minutes. Wash with TBS and detect according to preferred detection methods. With high sensitivity detection systems, typical working dilutions for the p73 antibody are 1:5,000 - 1;10,000.

      Optimal working dilutions must be determined by end user.
