      Applications
ApplicationDetect Myelin Proteolipid Protein using this Anti-Myelin Proteolipid Protein Antibody, C-terminus, clone PLPC1 validated for use in IH, IH(P) & WB.
      Key Applications
      • Immunohistochemistry
      • Immunohistochemistry (Paraffin)
      • Western Blotting
Application NotesWestern Blotting: detects a 23-25kDa protein and DM20 a 20kDa protein; PLP can aggregate causing HMW bands to sometimes appear in certain preparations.

Immunocytochemistry: 4% PFA fixed cells, 5-10 minutes room temperature. Use 0.1% triton in block only to allow antibody access to C-terminal epitope.

Immunohistology on fresh frozen sections or 4% PFA fixed, frozen tissues. Epitope is internal, use 0.1-0.2% triton X-100 in the blocking step for permeabilization.

Immunohistochemistry in paraffin: works best with lightly fixed tissues, or Bouin's fixed material, although there are reports of traditional formalin fixation also working with antigen recovery. {Mathis, et al 2003}.

Optimal working dilutions must be determined by end user.

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes a transmembrane proteolipid protein that is the predominant myelin protein present in the central nervous system. It may play a role in the compaction, stabilization, and maintenance of myelin sheaths, as well as in oligodendrocyte development and axonal survival. Mutations in this gene cause X-linked Pelizaeus-Merzbacher disease and spastic paraplegia type 2. Alternatively spliced transcript variants encoding distinct isoforms or having different 5' UTRs, have been identified for this gene.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P60201 # This is the major myelin protein from the central nervous system. It plays an important role in the formation or maintenance of the multilamellar structure of myelin.
SIZE: 277 amino acids; 30,077 Da
SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.
      PTM: Lipoprotein, Palmitate
DISEASE:Defects in PLP1 are the cause of Pelizaeus-Merzbacher disease (PMD) [MIM:312080]. PMD is an X-linked neurologic disorder of myelin metabolism. It is characterized by early impairment of motor development (during the first three months of life) and, later, by the development of abnormal movements and progressive spastic paraplegia.
SIMILARITY: Belongs to the myelin proteolipid protein family.
