      Background InformationInterleukin-6 (IL-6) is a pro-inflammatory cytokine that seems to be involved in the late stages of CTL generation. Other biological functions also include amplification and the inflammatory response. Combination therapies including IL-6 have been shown to have positive effects on specific malignant tumor types. The accumulation of IL-6 in the CNS is usually detrimental and can contribute to pathology of CNS diseases. IL-6 signaling is negatively regulated by TGF-beta, which down regulates STAT1, STAT3 and ICAM-1 expression. IL-6 expression has been linked to inflammatory bowel disease (IBD) and is also the primary clinical target for IBD. Clinical use of an antibody for the IL-6 receptor (IL-6R) has been promising in treating rheumatoid arthritis, systemic juvenile idiopathic arthritis and Castleman’s disease.

      UniProt SummaryFUNCTION: Cytokine with a wide variety of biological functions. It is a potent inducer of the acute phase response. Plays an essential role in the final differentiation of B-cells into Ig-secreting cells Involved in lymphocyte and monocyte differentiation. It induces myeloma and plasmacytoma growth and induces nerve cells differentiation Acts on B-cells, T-cells, hepatocytes, hematopoeitic progenitor cells and cells of the CNS. Also acts as a myokine. It is discharged into the bloodstream after muscle contraction and acts to increase the breakdown of fats and to improve insulin resistance.

      SUBCELLULAR LOCATION: Secreted.

      PTM: N- and O-glycosylated.

      POLYMORPHISM: Genetic variations in IL6 may be correlated with bone mineral density (BMD). Low BMD is a risk factor for osteoporotic fracture. Osteoporosis is characterized by reduced bone mineral density, disrutption of bone microarchitecture, and the alteration of the amount and variety of non-collagenous proteins in bone. Osteoporotic bones are more at risk of fracture.

      INVOLVEMENT IN DISEASE: Genetic variations in IL6 are associated with susceptibility to rheumatoid arthritis systemic juvenile (RASJ) [MIM:604302]. An inflammatory articular disorder with systemic-onset beginning before the age of 16. It represents a subgroup of juvenile arthritis associated with severe extraarticular features and occasionally fatal complications. During active phases of the disorder, patients display a typical daily spiking fever, an evanescent macular rash, lymphadenopathy, hepatosplenomegaly, serositis, myalgia and arthritis.

      Note=A IL6 promoter polymorphism is associated with a lifetime risk of development of Kaposi sarcoma in HIV-infected men.
