      Application CommentsBlocks IGF-I binding to the IGF-I receptor. May also weakly cross-react with the insulin receptor (see application references). It also inhibits the growth of MCF-7 cells in culture suggesting the IGF-I receptor may be involved in autocrine regulation of cell growth (see application references). Antibody should be titrated for optimal results in individual systems.

      Special InstructionsReconstitute with sterile phosphate buffered saline (PBS), pH 7.4, or sterile 20 mM Tris-saline (20 mM Tris containing 0.15 M NaCl), pH 7.4, to a final concentration of 100 µg/ml. Lyophilized antibodies should be resuspended at 4°C with occasional gentle mixing for at least two h. Following reconstitution, aliquot and freeze (-20°C) or refrigerate (4°C) with 0.1% sodium azide. Freezing of aliquots is recommended for long term storage of reconstituted product. Avoid freeze/thaw cycles of solutions.

      DescriptionPurified mouse monoclonal antibody generated by immunizing SJL mice with the specified immunogen and fusing splenocytes with FO mouse myeloma cells (see application references). Recognizes the ~130 kDa α and the ~90 kDa β subunits of IGF-I receptor.
      BackgroundThe insulin-like growth factors (IGFs) or somatomedins are mitogenic peptides present in the plasma of human, rat, and other species. Two distinct but chemically related IGFs have been purified from human plasma: IGF-I (Somatomedin C) and IGF-II. They are single chain polypeptides with molecular weights of 7,640 and 7,471, respectively. Types I and II IGF receptors differ in their structure and in their peptide binding specificity. Type I IGF receptors are structurally homologous to insulin receptors and contain a-subunits (MW = 130,000) that bind peptide, and b-subunits (MW = 90,000), which have intrinsic tyrosine kinase activity. The protein-tyrosine kinase activity of the insulin receptor and, presumably the IGF-I receptor, is required for insulin to stimulate all of its biological responses in cells. Type II receptors are a single polypeptide (MW = 260,000), lack protein-tyrosine kinase activity, and have been shown to be identical to the mannose-6-phosphate receptor.
