      Background InformationGap junction alpha-5 protein (UniProt: Q01231; also known as Connexin-40, Cx40) is encoded by the Gja5 (also known as Cxn-40) gene (Gene ID: 14613) in murine species. Gap junctions play an important role in a variety of cellular processes including homeostasis, intercellular metabolic cooperation and synchronization, morphogenesis, cell differentiation, and growth control. Connexins are tetraspan membrane proteins that form interlocking hexamers at each cell membrane to create a pore between two adjacent cells. Connexins usually have a short half-life, which is a key element of coupling regulation as it allows very dynamic and acute changes in gap junction regulation and gap junctional communication. Three types of connexins have been identified: Connexin 40, 43, and 45. Each of these connexins form channels with unique and specific electrophysiological properties. Connexin-40 is a multi-pass membrane protein with three cytoplasmic domains, four transmembrane domains, and two extracellular domains. Its expression is observed in kidney, heart, and skin; however, it is abundantly present in the lung tissue. It is also recognized as a component of gap junctions in vascular endothelium. It is reported to form channels with a large single channel conductance and moderate voltage-sensitivity. It is widely distributed in developing mouse heart and is expressed in the ventricles and atria at E11, however by day 14 it is restricted to the conduction system in the ventricles, but still, it remains in working atrial myocardium.
