      Background InformationCollagen a2(IV) chain (UniProt: P08572) is encoded by the COL4A2 gene (Gene ID: 1284) in human. Collagen, the most abundant protein in the body is predominantly synthesized by fibroblasts; however, epithelial cells may also synthesize small amounts of collagen. The collagen super family includes over 20 different types of collagen with at least 38 distinct polypeptide chains. Of these types I, II, and III are the most abundant and are involved in the formation of fibrils. Type IV collagen forms a two-dimensional reticulum and is a major component of the basal lamina. It is a network-forming collagen that underlies epithelial and endothelial cells and functions as a barrier between tissue compartments. Type IV collagen has many binding partners and forms the backbone of the basement membrane. The type IV collagen family of proteins comprises six isomeric chains, designated 1(IV) to 6(IV), each of which can form a triple helix structure with 2 other chains to generate type IV collagen network. Collagen IV alpha-2 contains a N-terminal collagenous 7S domain (aa 26-183), a central major collagenous domain that has over 20 interruptions of repeated -Gly-Xaa-Yaa- sequences, and the C-terminal non-collagenous (NC1) domain (aa 1489-1712). Each type of collagen IV is a heterotrimer composed of three alpha chains. Formation of these heterotrimers is initiated by the NC1 domain interactions and is accompanied by folding of the collagenous domains into triple helices. Type IV collagens also contain numerous cysteine residues that are involved in inter- and intramolecular disulfide bonding. Mutations in COL4A2 gene are known to cause brain small vessel disease that can lead to intracranial hemorrhage, fluid-filled cysts within the cerebral hemisphere, and delayed psychomotor development.
