      Background InformationCD9 antigen (UniProt: P40240; also known as CD9) is encoded by the Cd9 gene (Gene ID: 12527) in murine species. CD9 is a multi-pass membrane protein that belongs to the tetraspanin superfamily. Like other members of this family, CD9 has short N- and C- intracellular terminal tails and 4 transmembrane domains (aa 13-33; 54-74; 86-109; and 194-219) separating 2 extracellular domains (aa 34-53 and 110-193) of unequal size. CD9 is expressed predominantly in the peripheral nervous system and high expression is also observed in oocytes and blastocysts. It associates with integrins and is involved in the regulation of a variety of processes, including sperm-egg fusion, platelet activation and aggregation, and cell adhesion. It is reported to be present at the cell surface of oocytes where it plays a key role in sperm-egg fusion, possibly by organizing multiprotein complexes and the morphology of the membrane required for the fusion. CD9-/- female mice exhibit severely reduced fertility and CD9-/- oocytes also fail to fuse in vitro. Lower expression of CD9 has been linked with weaker matrix adhesion and diffuse tumor cell growth in vitro. Mice lacking CD9 exhibits signs of abnormal muscle regeneration and form discrete giant dystrophic myofibers. (Ref.: Rubenstein, E., et al. (2006). Dev. Biol. 290(2); 351-358; Charrin, S., et al.
