      Background InformationTBC1 domain family member 1 (UniProt: Q86TI0; also known as TBC1D1) is encoded by the TBC1D1 gene (Gene ID: 23216) in human. TBC1D1 is a member of the TBC1 Rab-GTPase family of nuclear proteins that is highly expressed in skeletal muscle. Its major action is to regulate exercise-mediated glucose uptake into muscle. It is involved in the trafficking and translocation of GLUT4-containing vesicles and insulin-stimulated glucose uptake into cells. Insulin and muscle contraction are reported to increase TBC1D1 phosphorylation on phospho-Akt substrate motifs. Insulin-stimulated phosphorylation by AKT is shown to stimulate GLUT4 translocation. Following muscle exercise, phosphorylation of TBC1D1 at serine 237 by AMPK and 14-3-3 protein binding capacity are significantly increased. Knockdown of TBC1D1 in C2C12 muscle cells is shown to increase fatty acid oxidation and reduce muscle levels of GLUT4. Although TBC1D1 knockout mice do not exhibit any disruption in their whole-body insulin action, they display impaired exercise-regulated metabolism.
