      Background InformationNibrin (UniProt O60934; also known as Cell cycle regulatory protein p95, Nijmegen breakage syndrome protein 1, p95 protein of the MRE11/RAD50 complex) is encoded by the NBN (also known as AT-V1, AT-V2, ATV, NBS, NBS1, P95) gene (Gene ID 4683) in human. Nibrin is a component of the MRN (MRE11/RAD50/NBN) complex involved in early steps of DNA double strand breaks (DSBs). The phosphorylation of H2AX pSer139 (γ-H2AX) found at the DNA-damaged sites, for example, is mediated by a direct interaction between Nibrin and the phosphorylated mediator of the DNA damage checkpoint 1 (MDC1). ATM, CtIP/RBBP8, Tip60, BRCA1, and SMC1 are also known interaction partners of Nibrin. Nibrin contains a fork-head associated (FHA) domain and two BRCA1 C-terminal (BRCT) tandem domains in its N-terminal region, followed by two MRE11-binding motifs and an ATM-binding motif in its C-terminal region. The Ser278 and Ser343 residues are known to be targeted for phosphorylation by ATM in response to ionizing-radiation (IR). NBN gene mutations are responsible for Nijmegen breakage syndrome (NBS), a disorder characterized by chromosomal instability, radiation sensitivity, microcephaly, growth retardation, immunodeficiency and predisposition to cancer.
