Background InformationNeurogenins are a family of bHLH transcription factors involved in specifying neuronal differentiation. They are related to Drosophila atonal. The neurogenins (ngns) make up one of these atonal-related gene families. In neural crest cells, the atonal-related neurogenin family is particularly important for the sensory lineage (Neurogenins Are Essential for the Formation of dorsal root ganglia), while the achaete-scute homologue ash1 (Mash1) is important for aspects of autonomic neurogenesis (Mash1 Is Essential for Noradrenergic Differentiation). Neurogenin-3 promotes pancreatic development.

      Entrez Gene Number
Entrez Gene SummaryNeurogenin-3 (NEUROG3) is expressed in endocrine progenitor cells and is required for endocrine cell development in the pancreas and intestine (Wang et al., 2006 [PubMed 16855267]). It belongs to a family of basic helix-loop-helix transcription factors involved in the determination of neural precursor cells in the neuroectoderm (Gradwohl et al., 2000 [PubMed 10677506]).[supplied by OMIM]

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q9Y4Z2 # Involved in neurogenesis. Also required for the specification of a common precursor of the 4 pancreatic endocrine cell types (By similarity).
SIZE: 214 amino acids; 22969 Da
SUBUNIT: Efficient DNA binding requires dimerization with another bHLH protein (By similarity).
      SUBCELLULAR LOCATION: Nucleus (Potential).
DISEASE: SwissProt: Q9Y4Z2 # Defects in NEUROG3 are the cause of congenital malabsorptive diarrhea 4 (DIAR4) [MIM:610370]. DIAR4 is an autosomal recessive disorder characterized by generalized malabsorption and a paucity of enteroendocrine cells. Patients with congenital diarrhea generally present within the first weeks after birth with severe, life-threatening watery diarrhea that can be classified as either secretory or malabsorptive in nature. Routine clinical evaluation, including intestinal biopsy, may be used to further categorize the diarrhea according to the severity of the inflammation and to assess the integrity of the crypt- villus axis and the architecture of the epithelial layer. On the basis of both clinical and pathological characteristics, various specialized formulas may be used to minimize the diarrheal symptoms, although some children require extended periods of intravenous nutrition to sustain normal growth and development.
SIMILARITY: SwissProt: Q9Y4Z2 ## Contains 1 basic helix-loop-helix (bHLH) domain.
