      Background InformationJAKs are non-receptor tyrosine kinases which couple to cytokine receptors, and are activated by cytokines. JAK kinases (JAK1, JAK2, JAK3, Tyk2) have a kinase domain and a second, pseudokinase domain which is not enzymatically active. The principal effectors of JAKs are the STATs, which are direct activators of transcription, but JAKs also signal to the Ras-MAP Kinase pathway, and through Tec, to the PI3 Kinase pathway. JAK3 is required for signaling through cytokine receptors which share a common γ chain. JAK2 is required for signaling through a different subset of cytokine receptors, while JAK1 is essential for signaling from the IL-2, IL-6 and interferon receptors. The stress-activated protein kinase 1 (SAPK) family is also referred to as the jun N-terminal kinase family in light of the substrate preference of these serine/threonine kinases. In addition to regulating many biological processes including proliferation and apoptosis, the JNK/SAPK family has been implicated in many neurodegenerative diseases including Alzheimer disease, Parkinson's disease and Amyotrophic lateral sclerosis.
