      Background InformationFree fatty acid receptor 2 (UniProt: O15552; also known as G-protein coupled receptor 43) is encoded by the FFAR2 (also known as FFA2, GPCR43, GPR43) gene (Gene ID: 2867) in human. FFAR2 is a multi-pass membrane glycoprotein with four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. It is expressed at relatively high levels in peripheral blood leukocytes and at lower levels in the spleen. It is a G-protein-coupled receptor that is activated by short chain fatty acids (SCFAs) and plays a role in the regulation of whole-body energy homeostasis and in intestinal immunity. In omnivorous mammals, SCFAs containing less than 6 carbons, such as acetate, propionate, and butyrate, produced by the gut microbiome, are the most potent activators. These SCFAs not only serve as a source of energy, but also act as signaling molecules. The activation of this G-protein-coupled receptor leads to the formation of inositol 1,4,5-trisphosphate leading to the mobilization of intracellular calcium and phosphorylation of ERK1 and ERK2. It also plays a role in glucose homeostasis by regulating the secretion of GLP-1. It is reported to regulate whole-body energy homeostasis through adipogenesis regulating both differentiation and lipid storage of adipocytes. In parallel, FFAR2 also mediates the activation of the inflammatory chemokines and cytokines. FFAR2 is also involved in maintaining healthy gut microbiome and suppress intestinal carcinogenesis and its down modulation has been linked to colon cancers. (Ref.: Sivaprakasam, S., et al. (2016).
