      Background InformationCytokeratins are a subfamily of intermediate filament proteins and are characterized by a remarkable biochemical diversity, represented in epithelial tissues by at least 20 different polypeptides. They range in molecular weight from between 40 kDa and 68 kDa and isoelectric pH between 4.9 - 7.8, The individual cytokeratin polypeptides are designated 1-20. The various epithelia in the human body usually express cytokeratins which are not only characteristic of the type of epithelium, but also related to the degree of maturation or differentiation within an epithelium. Cytokeratin subtype expression patterns are used to an increasing extent in the distinction of different types of epithelial malignancies.

      Immunohistochemistry on fresh frozen or acetone fixed tissue sections. Formalin fixed, paraffin embedded tissues are not recommended for RGE53.

      Immunocytochemistry: (10μg/mL) Light fixation with 4% formalin, 2.5% PFA ( 5-10 minutes room temperature) or acetone. {See, Barrett, et al 2001} Permeabilization with 0.2% triton X-100 in blocking buffer if using formalin or PFA fixations.

      Flow cytometry: Acetone fixed/ethanol fixed cells {See Mezosi, E et al, 2002}.

      Optimal working dilutions must be determined by the end user.

      SpecificitySpecifically recognizes glandular epithelial cells from gastro-intestinal tract, respiratory tract and urogenetical tract. As well as endocrine and exocrine tissues and mesothelial cells. It is unreactive with stratified squamous epithelia (esophagus, epidermis, ectocervix). In transitional epithelium of the bladder RGE53 only reacts with the superficial (umbrella) cells. RGE53 can distinguish adenocarcinomas and mesotheliomas from squamous cell carcinoma and non-epithelial tumours. The antibody is directed against 45 kD cytokeratin protein (cytokeratin 18). Cross reactivity with; Keratin from most mammals, birds.
