      Biological Information
ImmunogenRabbit anti-Apaf1 (C-terminus) polyclonal antibody was raised against a peptide corresponding to amino acids 1158 to 1177 of human Apaf1 (Zou et al. 1997). The sequence of the immunogenic peptide differs from that of murine Apaf1 by one amino acid (Zou et al. 1997; Cecconi et al. 1998).

      HostRabbit
SpecificityApoptosis is related to many diseases and is induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. The mammalian homologues of the key cell death gene CED-4 in C. elegans has been identified recently from human and mouse and was designated Apaf1 (for apoptosis protease-activating factor 1; Zou et al. 1997; Cecconi et al. 1998). Apaf1 binds to cytochrome C (Apaf2) and caspase-9 (Apaf3), which leads to caspase-9 activation. Activated caspase-9 in turn cleaves and activates caspase-3 which is one of the key proteases, being responsible for the proteolytic cleavage of many key proteins in apoptosis (Li et al. 1997). Apaf1 can also associate with caspase-4 and caspase-8 (Hu et al. 1998). Apaf1 is ubiquitously expressed in human tissues (Zou et al. 1997).

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes a cytoplasmic protein that initiates apoptosis. This protein contains several copies of the WD-40 domain, a caspase recruitment domain (CARD), and an ATPase domain (NB-ARC). Upon binding cytochrome c and dATP, this protein forms an oligomeric apoptosome. The apoptosome binds and cleaves caspase 9 preproprotein, releasing its mature, activated form. Activated caspase 9 stimulates the subsequent caspase cascade that commits the cell to apoptosis. Alternative splicing results in several transcript variants encoding different isoforms.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: O14727 # Oligomeric Apaf-1 mediates the cytochrome c-dependent autocatalytic activation of pro-caspase-9 (Apaf-3), leading to the activation of caspase-3 and apoptosis. This activation requires ATP. Isoform 6 is less effective in inducing apoptosis.
SIZE: 1248 amino acids; 141840 Da
SUBUNIT: Monomer. Oligomerizes upon binding of cytochrome c and dATP. Oligomeric Apaf-1 and pro-caspase-9 bind to each other via their respective NH2-terminal CARD domains and consecutively mature caspase-9 is released from the complex. Pro-caspase-3 is recruited into the Apaf-1-pro-caspase-9 complex via interaction with pro-caspase-9. Interacts with APIP.
      SUBCELLULAR LOCATION: Cytoplasm.
TISSUE SPECIFICITY: Ubiquitous. Highest levels of expression in adult spleen and peripheral blood leukocytes, and in fetal brain, kidney and lung. Isoform 1 is expressed in heart, kidney and liver.
DOMAIN: SwissProt: O14727 The CARD domain mediates interaction with APIP.
SIMILARITY: Contains 1 CARD domain. & Contains 1 NB-ARC domain. & Contains 13 WD repeats.
