OverviewThe His6-tagged fusion protein of UbcH5b is charge and support ubiquitinylation in vitro. Unlike their GST-tagged counterparts, the His6-tagged UbcH5 family members all appear to form thiol ester conjugates with ubiquitin at a similar rate under similar conditions. The His-tagged version of this enzyme is not susceptible to self-ubiquitinylation. UbcH5b is highly homologous to the S. cerevisiae UbC4/5 family of E2 enzymes, demonstrated to be essential for degradation of regulatory and abnormal proteins. UbcH5b, together with UbcH5a and UbcH5c, are the most active class of E2 enzymes in cell extracts and are associated with regulation of a number of transcription factors including p53, NF-κB, and c-Fos.
