Each different complex controls a specific transition between two subsequent phases in the cell cycle. CDK7 is the catalytic subunit of the CDK-activating kinase (CAK) complex, a serine-threonine kinase. CAK activates the cyclin-associated kinases CDC2/CDK1, CDK2, CDK4 and CDK6 by threonine phosphorylation. CAK complexed to the core-TFIIH basal transcription factor activates RNA polymerase II by serine phosphorylation of the repetitive C-terminus domain (CTD) of its large subunit (POLR2A), allowing its escape from the promoter and elongation of the transcripts. Involved in cell cycle control and in RNA transcription by RNA polymerase II. Its expression and activity are constant throughout the cell cycle.

How is this complex produced?It is a coexpressed protein - the cells are hit with all three viruses and the complex is purified via the his-tagged CDK - there is no invitro reconstitution. We are not able to comment if there are higher-order dimers (one complex dimerising with another).
What type of buffer is best for lysis when attempting to preserve a protein?The best buffer is Buffer A. It is composed of 50mM Tris/HCl, pH 7.5, 1mM EGTA, 1mM EDTA, 1mM Sodium Orthovanadate (activated), 10mM Na B-glycerolphosphate, 50mM NaF, 5mM Na pyrophosphate, 1% w/v Triton X-100, 0.1% w/v 2-mercaptoethanol, 1ug/ml each of aprotinin, leupeptin and pepstatin, 1mM PMSF and 1mM microcystin (this is optional). QS with water. You may store Buffer A in aliquots at -20°C for 6 months.
