Applications
ApplicationDetect Prosurfactant Protein B using this Anti-Prosurfactant Protein B Antibody, reacts with both C-terminus & N-terminus validated for use in ELISA, IH & WB.
Key Applications
  • ELISA
  • Immunohistochemistry
  • Western Blotting
Application NotesImmunohistochemistry: 1:4,000-1:8,000 on human tissue with and without antigen retrieval (see protocol on back) and 1:1,000-1:2,000 on adult and fetal mouse lung (see notes on back).

Immunoblot: 1:1,000-1:5,000; proprotein 42-46kDa.

ELISA: 1:500-1:1,000

Optimal dilutions must be determined by the end user.
Biological Information
ImmunogenFull length recombinant peptide (381 a.a.) of the human SP-B (pre)proprotein.
Epitopereacts with both C-terminus and N-terminus
HostRabbit
SpecificityReacts strongly with human and mouse proSP-B (detects both the N-terminal and C-terminal propeptides). Does not react with mature SP-B. On human tissue immunostains alveolar Type II cells and non-ciliated bronchiolar cells. On mouse tissue immunostains alveolar Type II cells and non-ciliated bronchiolar (Clara cells) in distal and terminal airways.

Entrez Gene Number
Entrez Gene SummaryThe SFTPB gene encodes the pulmonary-associated surfactant B protein (SPB), an amphipathic surfactant protein essential for lung function and homeostasis after birth. Pulmonary surfactant is a lipid-rich material that prevents lung collapse by lowering surface tension at the air-liquid interface in the alveoli of lung. SPB enhances the rate of spreading and increases the stability of surfactant monolayers in vitro. Surfactant is composed of phospholipids and other surfactant-associated proteins (Clark et al., 1995 [PubMed 7644495]). See also SFTPA1 (MIM 178630), SFTPC (MIM 178620), and SFTPD (MIM 178635).[supplied by OMIM]

UniProt Number
UniProt SummaryFUNCTION: SwissProt: P07988 # Pulmonary surfactant-associated proteins promote alveolar stability by lowering the surface tension at the air- liquid interface in the peripheral air spaces. SP-B increases the collapse pressure of palmitic acid to nearly 70 millinewtons per meter.
SIZE: 381 amino acids; 42117 Da
SUBUNIT: Homodimer; disulfide-linked.
SUBCELLULAR LOCATION: Secreted, extracellular space.
DISEASE: SwissProt: P07988 # Defects in SFTPB are the cause of pulmonary surfactant metabolism dysfunction type 1 (SMDP1) [MIM:265120]; also called pulmonary alveolar proteinosis due to surfactant protein B deficiency. Inborn errors of pulmonary surfactant metabolism are genetically heterogeneous disorders resulting in severe respiratory insufficiency or failure in full-term neonates or infants. These disorders are associated with various pathologic entities, including pulmonary alveolar proteinosis (PAP), desquamative interstitial pneumonitis (DIP), or cellular non- specific interstitial pneumonitis (NSIP).
SIMILARITY: SwissProt: P07988 ## Contains 1 saposin A-type domain. & Contains 3 saposin B-type domains.
MISCELLANEOUS: Pulmonary surfactant consists of 90% lipid and 10% protein. There are 4 surfactant-associated proteins: 2 collagenous, carbohydrate-binding glycoproteins (SP-A and SP-D) and 2 small hydrophobic proteins (SP-B and SP-C).
