  • Ig superfamily receptor LNIR
  • nectin 4
  • poliovirus receptor-related protein 4
  • nectin-4
  • Nectin-4
Background InformationNectin-4 is also known as poliovirus receptor-related protein 4 (PRR4 or PVRL4). Nectins are known for being the receptors for the herpes simplex virus. Nectin-4 is an immunoglobulin-like transmembrane cell adhesion protein that also seems to play a role in neural tube formation, with N-cadherin. Nectin-4 is connected with the actin cytoskeleton through afadin inside the cell. Nectin-4 is predominantly found in human placenta although in mice, expression is wide-ranging. Nectin-4 seems to adhere primarily to cells expressing nectin-1 and vice versa.

Entrez Gene Number
Entrez Gene SummaryPoliovirus receptor-like proteins (PVRLs), such as PVRL4, are adhesion receptors of the immunoglobulin superfamily and function in cell-cell adhesion (Reymond et al., 2001 [PubMed 11544254]).[supplied by OMIM].
Gene Symbol
  • PVRL4
  • PRR4
  • LNIR
Purification MethodProtein G Purified
UniProt Number
UniProt SummaryFUNCTION: Seems to be involved in cell adhesion through trans-homophilic and -heterophilic interactions, the latter including specifically interactions with PVRL2/nectin-1. Does not act as receptor for alpha-herpesvirus entry into cells.

SUBUNIT STRUCTURE: Self-associates. Interacts via its Ig-like V-type domain with PVRL1/nectin-1 Ig-like V-type domain. Interacts via its C-terminus with MLLT4.

SUBCELLULAR LOCATION: Cell membrane; Single-pass type I membrane protein Potential. Cell junction › adherens junction. Note: Colocalizes with MLLT4 at cadherin-based adherens junctions.

Processed poliovirus receptor-related protein 4: Secreted. Note: The secreted form is found in breast tumor patients.

TISSUE SPECIFICTY: Predominantly expressed in placenta. Not detected in normal breast epithelium but expressed in breast carcinoma.

PTM: The soluble form is produced by proteolytic cleavage at the cell surface (shedding), probably by ADAM17/TACE.

SEQUENCE SIMILARITIES: Belongs to the nectin family.

Contains 2 Ig-like C2-type (immunoglobulin-like) domains.

Contains 1 Ig-like V-type (immunoglobulin-like) domain.
