Background InformationEnhancer of filamentation 1, also known as CRK-associated substrate-related protein (CAS-L) or Cas scaffolding protein family member 2 or Neural precursor cell expressed developmentally down-regulated protein 9 (NEDD-9), or Renal carcinoma antigen NY-REN-12 p105, and encoded by the gene NEDD9 or CASL or CASS2, is a docking protein that plays a central role in coordinating tyrosine kinase mediated signaling for cell adhesion. NEDD9 controls the signals between focal adhesions at the cell periphery and the mitotic spindle in response to growth factors and signals. NEDD9 interacts with integrins and various other cellular proteins including BCAR3 and SH2D3C. It is localized to the cytoplasm in most cells but location can vary depending upon the cell type and function. NEDD9 is also found in cellular projections and lamellipodium and is found in the nucleus depending upon the cellular microenvironment. NEDD9 has multiple isoforms and active proteolytic fragments ranging from 55 kDa to 115 kDa. Isoforms p105 and p115 are predominantly cytoplasmic and associate with focal adhesions while p55 associates with the mitotic spindle. NEDD9 is highly expressed in most cells and tissues and is typically induced upon cell growth and down regulated in adult tissues such as brain. NEDD9 is processed during the cell cycle into multiple proteolytic fragments that appear to have important functions at specific times during the cell cycle process. In cancer NEDD9 over expression can promote tumor growth and NEDD9 acts directly to stabilize Aurora A kinase which is often over expressed in many cancers. Destabilization of Aurora A via the inhibition or destruction of NEDD9 allows for tumor reduction, thus NEDD9 expression is being examined as a potential cancer marker.
