Applications
ApplicationAnti-Integrin α4 Antibody, cytoplasmic on C-terminus is an antibody against Integrin α4 for use in ELISA, IP, RIA, WB, IH.
Key Applications
  • ELISA
  • Immunoprecipitation
  • Radioimmunoassay
  • Western Blotting
  • Immunohistochemistry
Application NotesELISA/RIA: 1:1000.

Western blot: 1:1000, non-reduced conditions; blocking should be with 1-3% BSA. Recognizes a 140 kDa human alpha 4 band under non-reducing conditions.

Immunohistochemistry: 1:500

Immunoprecipitation: 5 μL of antibody is sufficient to precipitate alpha4 from 5x106 cells.

Optimal working dilutions must be determined by end user.
Biological Information
ImmunogenSynthetic peptide derived from the COOH terminal sequence (cytoplasmic domain) of the human alpha 4 integrin subunit.
Epitopecytoplasmic domain, C-terminus
HostRabbit
SpecificityIntegrin alpha 4. Specificity determined by immunoprecipitation from a detergent extract of 35S methionine-labeled lymphoid cells. No cross-reactivity to alpha 1, alpha 2, alpha 3, alpha 5, alpha 6 or alpha V. The antibody recognizes an epitope on the cytoplasmic domain of the protein, and it is thus intracellular.

Entrez Gene Number
Entrez Gene SummaryThe product of this gene belongs to the integrin alpha chain family of proteins. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. This gene encodes an alpha 4 chain. Unlike other integrin alpha chains, alpha 4 neither contains an I-domain, nor undergoes disulfide-linked cleavage. Alpha 4 chain associates with either beta 1 chain or beta 7 chain.
Gene Symbol
  • ITGA4
  • MGC90518

UniProt Number
UniProt SummaryFUNCTION: SwissProt: P13612 # Integrins alpha-4/beta-1 (VLA-4) and alpha-4/beta-7 are receptors for fibronectin. They recognize one or more domains within the alternatively spliced CS-1 and CS-5 regions of fibronectin. They are also receptors for VCAM1. Integrin alpha- 4/beta-1 recognizes the sequence Q-I-D-S in VCAM1. Integrin alpha- 4/beta-7 is also a receptor for MADCAM1. It recognizes the sequence L-D-T in MADCAM1. On activated endothelial cells integrin VLA-4 triggers homotypic aggregation for most VLA-4-positive leukocyte cell lines. It may also participate in cytolytic T-cell interactions with target cells.
SIZE: 1038 amino acids; 115333 Da
SUBUNIT: Heterodimer of an alpha and a beta subunit. The alpha subunit can sometimes be cleaved into two non-covalently associated fragments. Alpha-4 associates with either beta-1 or beta-7. Alpha-4 interacts with PXN, LPXN, and TGFB1I1/HIC5. Interacts with CSPG4 through CSPG4 chondroitin sulfate glycosaminoglycan.
SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein.
DOMAIN: SwissProt: P13612 The SG1 motif is involved in binding to chondroitin sulfate glycosaminoglycan and cell adhesion.
PTM: Phosphorylation on Ser-1027 inhibits PXN binding.
SIMILARITY: Belongs to the integrin alpha chain family. & Contains 7 FG-GAP repeats.
