Applications
ApplicationDetect Dopamine D2 Receptor using this Anti-Dopamine D2 Receptor Antibody, cytoplasmic domain, long form validated for use in ELISA, IH, IP & WB.
Key Applications
  • ELISA
  • Immunohistochemistry
  • Immunoprecipitation
  • Western Blotting
Application NotesWestern blot: 1-10 μg/mL (Chemiluminescence technique) The predicted size of D2L form is approximately 50 kDa. However, additional bands at approximately 60 and 100 kDa, and others are typically seen in various tissues (Boundy, et al. 1993; Sakata, et al. 1992; Farooqui et al. 1991, 1992; Brock et al. 1992; Farooqui & Prasad 1992). Approximately 100-400 μg of membrane protein/lane should be loaded to visualize various bands.

Immunohistochemistry: 1-20 μg/mL on paraformaldehyde fixed sections.

Immunoprecipitation: 1-10 μg using 100-400 μg of protein (Boundy, et al. 1993).

ELISA: 0.5-1.0 μg/mL (1 μg/mL immunogen peptide (AG211)/well)

Optimal working dilutions must be determined by the end user.
Biological Information
ImmunogenA 27 amino acid peptide sequence from the rat D2 long form within the cytoplasmic loop #3.
Epitopecytoplasmic domain, long form
HostRabbit
SpecificitySpecific for Dopamine Receptor D2 long form. Dopamine D2 receptor is known to exist in the short (D2S) and the long (D2L) forms that are encoded by splice variants of a single gene and differ only by the presence of an additional 29 amino acid in the intracellular, cytoplasmic loop 3 (Dal Toso et al. 1989; Grandy et al. 1989; Monsma et al. 1990; Giros et al. 1989). D2L receptor is a 446 aa, G-protein coupled and transmembrane receptor protein (Boundy et al. 1993) The 27 aa rat D2RL peptide used for immunization was found only in the D2 long form and there is no significant homology with other dopamine receptors (D1, D2 short, and D3-D5).

SPECIES REACTIVITIES: The immunogen peptide shows sequence homology of 96% with human/bovine (26/27 aa), 62% with Xenopus (17/27 aa) D2L receptors. Antibodies generated to the rat sequence have been shown to recognize the canine D2R (Boundy et al. 1993). Antibody cross-reactivity with various species is not known.

Entrez Gene Number
Entrez Gene SummaryThis gene encodes the D2 subtype of the dopamine receptor. This G-protein coupled receptor inhibits adenylyl cyclase activity. A missense mutation in this gene causes myoclonus dystonia; other mutations have been associated with schizophrenia. Alternative splicing of this gene results in two transcript variants encoding different isoforms. A third variant has been described, but it has not been determined whether this form is normal or due to aberrant splicing.

  • D2R
UniProt Number
UniProt SummaryFUNCTION: SwissProt: P14416 # This is one of the five types (D1 to D5) of receptors for dopamine. The activity of this receptor is mediated by G proteins which inhibit adenylyl cyclase.
SIZE: 443 amino acids; 50619 Da
SUBUNIT: Interacts with GPRASP1, neurabin-2 and CLIC6 (By similarity). Interacts with CADPS and CADPS2.
SUBCELLULAR LOCATION: Cell membrane; Multi-pass membrane protein.
DISEASE: SwissProt: P14416 # It has been suggested that DRD2 is involved in psychiatric disorders; especially in schizophrenia. & Defects in DRD2 are associated with myoclonus dystonia (MD) [MIM:159900]; also known as myoclonic dystonia, alcohol- responsive dystonia and dystonia-11 (DYT11). MD is a movement disorder characterized by involuntary lightning jerks and dystonic movements and postures alleviated by alcohol. Inheritance is autosomal dominant. The age of onset, pattern of body involvement, presence of myoclonus and response to alcohol are all variable.
SIMILARITY: SwissProt: P14416 ## Belongs to the G-protein coupled receptor 1 family.
