DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with Sp2/0-Ag14 mouse myeloma cells. Recognizes the ~180 kDa clathrin heavy chain protein.
BackgroundAt the unstimulated presynaptic nerve terminal, a pool of synaptic vesicles remains docked at active zones in preparation for exocytosis. Nerve terminal depolarization induced through an action potential stimulates the rise in intracellular Ca2+, which ultimately initiates the exocytotic process. Fused synaptic vesicles are rapidly replaced with vesicles from the reserve pool, while membranes of the fused vesicles are recaptured by endocytosis. Clathrin is one component involved in vesicle endocytosis, and is composed of three polypeptides, a ~180 kDa heavy chain and two ~32-38 kDa light chains which combine to create a distinct three-legged triskelion. It is this morphology that allows clathrin to form its unique polyhedral network. The clathrin-adaptor AP2 and the AP180/AP3 proteins also play a role in this process by forming a protein layer between the membrane and clathrin lattice. The interaction of the AP2 heterotetramer with synaptotagmin is believed to act as the template for recruitment of the AP3 and clathrin triskelia complex. Progression from clathrin-coated to free vesicles in the nerve terminus also requires dynamin.
