Merck:/Freestyle/BI-Bioscience/Antibodies-Assays/neuroscience-images/neuro_icon10_round.gifOxidative stress is a cellular disorder caused by an excess of free radicals resulting from increased oxidant levels, decreased anti-oxidant amounts, or failure to repair damage induced by reactive oxygen species (ROS). Oxidative stress and oxidation have been linked to atherosclerosis, ischemia, cancer, rheumatoid arthritis, and neurodegeneration. ROS molecules can chemically modify proteins, lipids, and nucleic acids, potentially altering or impairing their normal functions.

Protein oxidation is frequently associated with excess oxidative stress in the cells. Oxidative modification of proteins, by oxygen free radicals and other reactive species such as hydroxynonenal, occurs in physiologic and pathologic processes. As a consequence of the modification, carbonyl groups are introduced into protein side chains by a site-specific mechanism.

Merck’s offers a broad range of products for studying oxidative stress, including detection kits, multiplex assays, flow cytometry characterization, antibodies, and inhibitors. Our oxidative stress detection kits enable simple and sensitive immunodetection of carbonyl-modified proteins, and allow reliable and reproducible quantitation.

Oxidative Stress, Carbonyl Modification Detection
Oxidative stress introduces carbonyl groups in proteins. These can be chemically modified (DNP-derivatized) and subsequent detected with antibodies specific for DNP, and using various applications.

DNP in serial dilutions of DNP-protein mixture provided in the kit were analyzed as described in the protocol. Exposure time was 1 minute. DNP in BSA Band: lane 1: 100 femtomoles,  lane 2: 30 femtomoles,  lane 3: 10 femtomoles, lane 4: 3 femtomoles, lane 5: 1 femtomoles, lane 6: 0.3 femtomoles, lane 7: 0.1 femtomoles

OxyICC™ Oxidized Protein Detection Kit

The OxyICC™ Oxidized Protein Detection Kit provides reagents for fluorescent immunocytochemical detection of cellular protein carbonyls. This simple assay detects carbonyl modifications using dinitrophenylhydrazine (DNPH) to provide highly sensitive and quantitative results.
