      Detection of mouse VEGFR2, phosphorylated at Tyr1054/1059, by immunoblotting. Samples: Whole cell lysate from porcine aortic endothelial (PAE) cells transfected with CSF-1/mouse VEGFR2 chimeric receptor left unstimulated (lanes 1,2) or stimulated with CSF-1 (lanes 3-8). Primary antibodies: PhosphoDetect™ Anti-VEGFR2 (pTyr1054/1059) Rabbit pAb (Cat. No. PS1013) (0.5 µg/ml) pre-incubated without peptide (lanes 2,4,5), with non-phosphopeptide corresponding to the immunogen (lane 6), a generic phosphotyrosine-containing peptide (lane 7), or the phosphopeptide immunogen (lane 8) or pan anti-VEGFR2 antibody (lanes 1,3). Detection: chemiluminescence.

      Application CommentsThe immunogen sequence is 100% conserved in human and rat VEGFR2, but reactivity has not been tested. The corresponding phosphorylation site in VEGFR3 is 100% conserved, but cross-reactivity has not been tested. When used at a 1:1000 dilution there is sufficient antibody for 10 immunoblots at 10 ml/blot. Antibody should be titrated for optimal results in individual systems.

      BackgroundVascular Endothelial Growth Factor Receptor 2 (VEGFR2) is receptor tyrosine kinase that serves as a key mediator in angiogenesis and plays an essential role in embryonic development, wound healing, cell proliferation, migration, and differentiation. It contains seven immunoglobulin-like sequences in the extracellular domain and exhibits sequence similarity to Flt-1 (FMS-like tyrosine kinase 1) and Flt-4 (FMS-like tyrosine kinase 4). Upon binding to its ligand, VEGF, VEGFR2 undergoes ligand-induced dimerization, which activates its intrinsic tyrosine kinase activity resulting in auto- and trans-phosphorylation on specific tyrosine residues in the cytoplasmic domain. These tyrosine residues, when phosphorylated, are involved as docking sites to recruit molecules containing Src homology 2 (SH2) or phosphotyrosine-binding domains and to convey signals to downstream effectors. Phosphorylation of VEGFR2 at Tyr1054 and Tyr1059 within the activation loop is required for required for activation of VEGFR2 and enhances its intrinsic tyrosine kinase activity. Phosphorylation at Tyr1059 is also responsible for VEGF-induced intracellular Ca2+ mobilization.
