      BackgroundThe nucleosome is composed of ~200 bp of DNA and eight histone proteins, two from each of the four histone protein families, H4, H3, H2B, and H2A. H2A includes three subfamilies: H2A1-H2A2, H2AZ, and H2AX, which have been conserved throughout eukaryotic evolution. Phosphorylation of H2AX on serine 139 by ATM or DNA-dependent protein kinase (DNA-PK) is an early step in the response to DNA damage by ionizing radiation. Phosphorylation of H2AX is required for the accumulation of 53BP1 at DNA break sites.
