      Recommended reaction conditions50 mM HEPES buffer, pH 6.5-7.0, 10 mM CaCl2, 0.5-1 mM 4,4'-dithiodipyridine, 0.01-5 mM substrate, and desired amount of activated enzyme; incubate at 25-26° while continuously monitoring cleavage at 324 nm.
