      Recommended reaction conditionsMaterials Required:Load/Equilibration Buffer: 20 mM sodium phosphate, pH 7.5, 150 mM NaCl • Wash Buffer: (same as Load/Equilibration buffer) • Elution Buffer: 20 mM sodium phosphate, pH 7.5, 150 mM NaCl, 100 mM N-Acetyl-α-D-glucosamine (GlcNAc, Cat. No. 1079) • Regeneration Buffer: (same as Load/Equilibration buffer) • Storage Buffer: 10 mM sodium phosphate, pH 7.5,150 mM NaCl, 0.05% NaN3 Protocol Note: Lectin column and negative control column should be used under identical conditions. 1. Buffers should be chilled to 4°C prior to use. Do not allow column to run dry. 2. Remove top and bottom caps from column. 3. Wash with 10 ml or more Equilibration Buffer. 4. Dissolve ligand in Equilibration Buffer to a concentration of ~2 mg/ml. Be sure that ligand is completely dissolved before proceeding. Clarify if necessary. 5. Apply ligand solution to column. 6. Wash with 20 ml or more Wash Buffer. Note: Some carbohydrates interact weakly with WGA. As a result, these carbohydrates are merely retarded by the immobilized WGA and will elute in the Wash Buffer. Wash Buffer fractions should be analyzed for the presence of carbohydrate. 7. Elute resin with a minimum of 5 ml Elution Buffer. 8. To reuse column, wash extensively (>20 ml) with Regeneration Buffer. 9. Wash column with 3 ml storage buffer. Store at 4°C.
