      Background InformationSyntaxin-1A (UniProt Q16623; also known as Neuron-specific antigen HPC-1) is encoded by the STX1A (also known as STX1, SYN1A) gene (Gene ID 6804) in human. Syntaxin-1A belongs to the superfamily of soluble N-ethylmaleimide-sensitive factor (NSF) attachment protein (SNAP) receptor (SNARE) proteins involved in mediating vesicle fusion events. Target or t-SNAREs located in the membranes of target compartments form stable subcomplexes and engage vesicle or v-SNAREs in the membranes of transport vesicles to facilitate the fusion. Syntaxin-1A is a t-SNARE abundantly expressed in most neurons and localized on the neuronal plasma membrane, where it participates in the formation of the trans-SNARE complex by engaging t-SNARE SNAP25 (synaptosomal-associated protein 25) and v-SNARE synaptobrevin (a.k.a. vesicle-associated membrane protein2 or VAMP2) to allow fusion between the synaptic vesicle and plasma membrane. Two types of syntaxin 1 isoforms exist. Studies utilizing Stx1a(-/-) and Stx1b(-/-) knockout mice indicate that these two syntaxin isoforms play distinct roles in neuronal survival.
