      Background InformationPolycystin-2 (UniProt: Q13563; also known as PC2, Autosomal dominant polycystic kidney disease type II protein, Polycystic kidney disease 2 protein, Polycystwin, R48321, Transient receptor potential cation channel subfamily P member 2) is encoded by the PKD2 (also known as TRPP2) gene (Gene ID: 5311) in human. Polycystin-2 is a multi-pass membrane protein that serves as a non-selective cation channel formed by PKD1 and PKD2 and is activated by interaction between PKD1 and a Wnt family member (WNT3A and WNT9B). It is also reported to form a functional, homotetrameric ion channel. It functions as a cation channel involved in fluid-flow mechanosensation by the primary cilium in renal epithelium. Its channel activity is regulated by phosphorylation and intracellular calcium levels. PKD-mediated phosphorylation at the C-terminus regulates its function in the release of Ca2+ stores from the endoplasmic reticulum. It acts mainly as an outward-rectifying K+ channel, but is also permeable to Ca2+, and to a much lesser degree to Na+. It is also shown to contribute to the release of Ca2+ stores from the endoplasmic reticulum. Mutation is PKD2 gene have been linked to polycystic kidney disease with or without polycystic liver disease that is characterized by progressive formation and enlargement of cysts in both kidneys, typically leading to end-stage renal disease in adult life.
