Background InformationCyclin-dependent kinase 11A (UniProt: Q9UQ88; also known as EC:2.7.11.22, Cell division cycle 2-like protein kinase 2, Cell division protein kinase 11A, Galactosyltransferase-associated protein kinase p58/GTA, PITSLRE serine/threonine-protein kinase CDC2L2) is encoded by the CDK11A (also known as CDC2L2, CDC2L3, PITSLREB) gene (Gene ID: 728642) in human. CDK11 is a ubiquitously expressed protein that plays multiple roles in cell cycle progression, cytokinesis, and apoptosis. Eight different isoforms of CDK11 have been described that are produced by alternative splicing and alternative initiation. The p110 isoform (canonical) is suggested to be involved in pre-mRNA splicing, potentially by phosphorylating the splicing protein SFRS7. During apoptosis, it is induced by Fas or tumor necrosis factor. The p110 isoform is cleaved by caspases to produce p110C form that contains the C-terminal kinase domain and binds to the serine/threonine kinase PAK1. CDK11 is reported to associate with SF3B1 and phosphorylates threonine residues at its N terminus during spliceosome activation. This phosphorylation event is shown to be essential for the association between SF3B1 and U5 and U6 snRNAs in the activated spliceosome, termed the Bact complex. CDK11 is overexpressed in several tumor types and is a predictor of poor clinical outcome. (Ref.: Blazek, D. (2023). Clin. Transl Med. 13(3); e1201; Hluchý, M., et al. (2022).
