      Product Description


      Bead-based flow cytometric assays enable sensitive, precise quantification of analytes within a sample. When directed toward proteins or peptides, such assays are essentially ELISAs on a bead. Samples are combined with fluorescently labeled microparticles (beads) covalently conjugated to a capture antibody. Analytes captured on the beads are identified with detection antibodies and a fluorescent label. A major advantage of bead-based assays over traditional protein quantification methods (such as ELISA) is the capacity for multiplexing, as bead-based assays allow simultaneous quantification of multiple analytes in a small sample volume.



      Panel Overview

      Heat shock proteins (HSPs) are a family of proteins that function as molecular chaperones within the cell, and have many important roles including folding nascent proteins and maintaining proteins in a folded, active state. They can be induced in response to a variety of stresses including heat shock, radiation, and cytotoxic drug exposure, and have increased expression in cancer cells relative to normal cells. A number of these proteins are being investigated as potential targets for cancer therapy, as they can have effects on proliferation, apoptosis, metastasis, and chemotherapy resistance in certain cancers. The levels of some of the family members are coordinately regulated by the transcription factor Heat Shock Factor 1 (HSF1), in response to a variety of cellular stresses. Interestingly, HSP90 inhibitors can disrupt the interaction of HSP90 with HSF1, which can lead to increased expression of HSP family members such as HSP70 and influence the efficacy of treatment. To better understand the roles and regulation of heat shock proteins and the heat shock response in cancer and other physiological processes, there is increased interest in simultaneous analysis of key heat shock proteins in a panel or multiplex format, rather than studying each protein in isolation.

      The BeadPlex® Human Heat Shock Protein (HSP) Panel is designed for the in vitro quantitative determination of the following analytes in human cell lysates and tissue extracts:



      •HSP27 (Total)
      •Phospho-HSP27(Ser78/Ser82)
      •HSP60
      •HSP70
      •HSP90α

      This assay is validated for human samples. Other species have not been tested. This kit has not been tested for multiplexing with other markers. Should the user elect to multiplex this kit with other Luminex® kits, the assay conditions should be determined empirically for each specific application.



      Panel Components

      Storage: All components of the BeadPlex Human HSP Panel are shipped at 2 to 8°C. Upon receipt, store all kit components at 2 to 8°C. Do not freeze.


      The BeadPlex Human HSP Panel is available in two kit configurations:
      1. Complete Kit
      2. Mix and Match Format.

      For end-user convenience, customers may order the BeadPlex Human HSP Panel - Complete Kit (Cat. No. BPHHSP-5). The Complete Kit contains a mixture of capture beads, detection antibodies, and enough reagents and buffers to assay 96-well plate of the aforementioned analytes.

      For maximum flexibility, end-users may configure their own customized multiplex assay through the Mix and Match Format. This format consists of a BeadPlex Human HSP - Base Kit (Cat. No. BPHHSP-BASE) and a customized choice of any or all panel specific capture beads as listed in Technical Bulletin (TB547). The Base Kit and selected components contain enough reagents and buffers to assay one 96-well plate.
