      Revision13-September-2008 RFH
SynonymsTNF-α Convertase Enzyme Substrate, Fluorogenic, ADAM-17 Substrate II, Fluorogenic, MCA-PLAQAV(Dpa)RSSSR-NH₂
DescriptionA fluorescence energy transfer (FRET) peptide substrate for tumor necrosis factor (TNF)-α convertase (TACE) (Km ~19 µM). Cleavage of this TACE substrate II occurs between the alanine and valine residues. Useful for the screening of TACE inhibitors. Excitation max: ~320 nm; emission max: ~395 nm.
      FormLiquid
FormulationA 4 mM (7.5 mg/ml) solution in DMSO.
Recommended reaction conditions50 mM Tricine buffer, pH 7.5, containing 100 mM NaCl, 10 mM CaCl2, and 1 mM ZnCl2, 10 µM TACE Substrate II, 600 nM TACE; incubate at 27°C with mechanical shaking; reaction can be quenched with 50 mM EDTA (final concentration); cleavage monitored at excitation at ~320 nm and emission at ~395 nm.
