      Background InformationHistone deacetylase 1 (UniProt: Q13547; also known as EC:3.5.1.98, HD1, HDAC1) is encoded by the HDAC1 (also known as RPD3L1) gene (Gene ID: 3065) in human. HDAC1 is a ubiquitous enzyme that is expressed at higher levels in heart, pancreas, and testis and lower levels in kidney and the brain. It is a part of the core histone deacetylase complex that is composed of HDAC1, HDAC2, RBBP4 and RBBP7. HDAC1 is responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4) and this deacetylation gives a tag for epigenetic repression. It plays an important role in transcriptional regulation, cell cycle progression, and developmental events. HDAC1 is also a component of the BRG1-RB1-HDAC1 complex that negatively regulates the CREST-mediated transcription in resting neurons. Upon calcium stimulation, HDAC1 is released from the complex and CREB binding protein is recruited, which facilitates transcriptional activation. Phosphorylation of HDAC1 is shown to stabilize its interactions with binding partners in multiprotein complexes. Phosphorylation on serine 421 and serine 423 are constitutively phosphorylated by casein kinase II to regulate its activity. Clone BT-15 specifically detects HDAC1 phosphorylated on serine 406 in the prophase state, which is critical for cells to prepare for mitosis and marks the onset of events for cell division.
