      Application Commentsc-erbB4 exists as two alternatively-spliced isoforms. Does not cross react with c-erbB1 (EGFR), c-erbB2/HER-2 or c-erbB3/HER-3. This antibody does not interfere with the receptor-heregulin binding and is particularly well suited for immunoprecipitation of unoccupied as well as the occupied c-erbB4 receptor. It reacts with intact cells, stimulates tyrosine kinase activity and efficiently stimulates internalization of c-erbB4 (use antibody at 10 µg/ml). This antibody has also been shown to work for flow cytometry. Antibody should be titrated for optimal results in individual systems.

      DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with mouse myeloma NSO cells (see application references. Recognizes the ~180-185 kDa erbB4/HER-4 protein.
      Backgroundc-erbB4/HER-4 is the fourth member of the type I transmembrane tyrosine kinase family of growth factor receptors that includes c-erbB1 (EGFR), an orphan receptor, c-erbB2 and c-erbB3, a receptor for the neu differentiation factor (NDF/heregulin). Like c-erbB3, c-erbB4 is also a receptor for NDF/heregulin. Each receptor member is characterized by ectodomains with two cysteine rich regions however, despite their structural homology, they differ in ligand specificities. c-erbB1/Her-1 binds EGF whereas c-erbB3 and c-erbB4 are the respective low and high affinity receptors for many isoforms of NDF/heregulin. Currently the ligand specificity for c-erbB2 has not been well characterized and thus remains the 'orphan receptor'. All four receptor members are expressed in breast cancer presumably a result of gene amplification. Overexpression of perhaps all of the receptors can also occur, probably as a result of increased mRNA transcription.
