      Background InformationH1.4 is one of four variants of histone H1 (H1.2-H1.5) that is found in somatic cells. The H1 histones may function as linker proteins binding DNA via their GH domain, and enabling the wrapping of DNA around the core nucleosomal structure formed from the association of H2A, H2B, H3, and H4. In response to various stimuli, histones undergo a number of post-translational modifications (PTM) including acetylation, phoshorylation, and methylation. Previous studies have suggested that H1.4 may be phosphorylated during mitosis and interphase resulting in increased transcription of target genes by RNA polymerase I and II, and therefore this PTM on H1.4 may have implications for cell growth. H1.4 may also undergo acetylation of its N-terminal lysine residues. Often, it is the pattern of different modifications on several histone residues that alter chromatin and affect gene expression, rather than modification of single residues.
