      Background InformationZika virus is a member of the Flaviviridae family that contains a single positive sense RNA genome. Following the fusion of virus membrane with host endosomal membrane, RNA genome is released into the cytoplasm. It is initially translated as a single polyprotein, which is post-translationally cleaved into three structural proteins (C, PrM or M, and E) and seven nonstructural (NS) proteins. Six of the NS proteins (NS2A to NS5) are shown to form a replication complex on the cytoplasmic side of the endoplasmic reticulum membrane. The glycosylated NS1 that is reported to associates with lipids, forms a homodimer inside the cells and is shown to be essential for viral replication and late in infection. It is also secreted into the extracellular space as a hexameric lipoprotein particle and plays a role in immune evasion and pathogenesis via its interaction with components from both the innate and adaptive immune systems. NS1 is also a major antigenic marker for viral infection. NS2A functions in virion assembly and antagonizes the host immune response. The non-structural protein 2B is essential for the serine protease function of NS3. The serine protease NS3 has three different enzymatic activities - serine protease, NTPase and RNA helicase. The non-structural protein 4A of virus regulates the ATPase activity of the NS3 helicase while the non-structural protein 4B inhibits interferon-induced host STAT1 phosphorylation and its nuclear translocation.
