      BackgroundFunctional mTOR (mammalian target of rapamycin) exists in cells in complex with other proteins, including Raptor and Rictor, which are two unrelated, cytosolic proteins that bind to and regulate mTOR. mTORC1 (mTOR complex 1) consists of mTOR, Raptor (regulatory associated protein of mTOR), and mLST8/GβL (mammalian LST8/G-protein β-subunit like protein) and functions as a nutrient/energy/redox sensor and controller of protein synthesis. Growth factor signaling, together with nutrient availability, regulates cell growth by controlling the activity of mTORC1. mTORC2 (mTOR complex 2) consists of mTOR, Rictor (rapamycin-insensitive companion of mTOR, acc # NP_689969.2), GβL, and mSIN1 (mammalian stress-activated protein kinase interacting protein 1) and functions as an important regulator of the cytoskeleton through its stimulation of F-actin stress fibers, paxillin, RhoA, Rac1, Cdc42, and PKCα. mTORC2 is also believed to regulate responses to cellular stress via the FOXO pathway. mTORC2 is also known to be the elusive PDK2, a serine/threonine kinase that phosphorylates Akt on Ser473. Biomarkers indicate that the mTOR pathway is overactive in certain cancers, suggesting that mTOR may be an attractive target for cancer therapy.
