      Background InformationMitotic checkpoint protein BUB3 (UniProt O43684; also known as Budding uninhibited by benomyl, Budding uninhibited by benzimidazoles 3 homolog, hBUB3) is encoded by the BUB3 (also known as BUB3L) gene (Gene ID 9184) in human. The mitotic checkpoint is activated when there is unattached kinetochores to spindle microtubules. When all kinetochores are attached to spindle microtubules and bi-orientation of sister chromatids is established, the mitotic checkpoint signaling is silenced and chromosome segregation proceeds. BUB3 is a key mitotic checkpoint factor that recognizes centromeric components and recruits other mitotic checkpoint molecules to the unattached kinetochore, where they assemble as an active complex. BUB3 interacts with BUB1, BubR1, MAD2, and CDC20 through its WD repeat, while the region 213-EVQKKKYAFK-222 is reported to be essential for proper BUB3 nuclear localization and its interaction with CENP-A and KNL1.
