      Background InformationMethylcytosine dioxygenase TET1 (UniProt: Q3URK3; also known as CXXC-type zinc finger protein 6, Ten-eleven translocation 1 gene protein homolog) is encoded by the Tet1 (also known as Cxxc6, Kiaa1676) gene (Gene ID: 52463) in murine species. TET1 is a dioxygenase that catalyzes the conversion of the modified genomic base 5-methylcytosine (5mC) into 5-hydroxymethylcytosine (5hmC) and plays a key role in active DNA demethylation. It is shown to be present in embryonic stem cells. Presence of 5hmC has been reported in the brain, especially in Purkinje neurons. In addition to its role in DNA demethylation, TET1 also plays a more general role in chromatin regulation. It preferentially binds to CpG-rich sequences at promoters of both transcriptionally active and Polycomb-repressed genes. It is involved in the recruitment of the O-GlcNAc transferase OGT to CpG-rich transcription start sites of active genes, thereby promoting histone H2B GlcNAcylation by OGT. TET family consists of three TET enzymes that contain cysteine rich and DSBH domains. TET1 and TET3 also have a CXXC domain, which typically bind unmethylated CpG dinucleotides, but in TET enzymes they bind 5mC and 5hmC.
