      Background InformationProtein sulfenic acids (SOHs) are the principal oxidation products formed when redox active proteins interact with peroxide molecules. Cysteine, a unique thiol containing amino acid, can readily undergo a range of oxidative post-translational modifications and sulfenic acid is a transient reaction intermediates of cysteine thiol. Sulfenic acid can act as an electrophile and react selectively with 1,3-diketones such as 5,5-dimethyl-1,3-cyclohexanedione (dimedone) under physiological conditions to generate corresponding thioester derivative. Cysteine sulfenic acid (Cys-SOH) is a common post-translational modification in proteins and reversible oxidation of Cys-SH to Cys-SOH is observed in several biological events. In the presence of molecular oxygen and trace metals, protein-SOH can form spontaneously in unfolded proteins, even in the absence of any added oxidants. Protein sulfenic acid formation can be used to monitor protein oxidation on reactive cysteines within target proteins. This rabbit polyclonal antibody recognizes dimedone-tagged sulfenic acid containing proteins and offers an immunochemical detection for sulfenic acid. This antibody has been used to show that the hyperoxidized glyceraldehyde 3-phosphate dehydrogenase (GAPDH) causes oxidation of its Cys-149 sulfenate to SO2 and SO3 form and inhibits glycolysis in ventricular myocytes in rats.
