      BackgroundCtBP interacting protein (CtIP; Accession No. NP_002885), originally identified as a binding partner of the transcriptional suppressor CtBP, was subsequently shown by two-hybrid screening to interact with BRCA1 BRCT domains. CtIP is a phosphorylation-dependent binding partner of the BRCA1 BRCT domains. The cell cycle-regulated BRCA1/CtIP complex is required for the G2/M transition and Chk1 activation following DNA damage. CtIP interacts with a group of tumor suppressor proteins including Rb (retinoblastoma protein), BRCA1, Ikaros, and CtBP, which regulate cell cycle progression through transcriptional repression as well as chromatin remodeling. CtIP associates with SIAH-1 both in vitro and in vivo leading to CtIP degradation by the ubiquitin-proteasome pathway. CtIP(-/-) embryos die at embryonic day 4 with blastocysts arrested in G1, in an Rb dependent manner. CtIP(+/-) heterozygotes are characterized by the development of multiple types of tumors, predominantly, large lymphomas. CtIP becomes hyperphosphorylated and dissociated from BRCA1 upon exposure to ionizing radiation. This phosphorylation event requires the protein kinase, ATM, that is mutated in the disease ataxia telangiectasia. ATM phosphorylates CtIP at Ser664 and Ser745. ATM modulates BRCA1-mediated regulation of the DNA damage-response GADD45 gene through CtIP, thus providing a potential link between ATM deficiency and breast cancer.
