DescriptionAnti-Calpain Antibody, small subunit of µ- or m-Calpains (Calpain I or II), clone P1
Background InformationCalpains are non-lysosomal cysteine proteeases which absolutely require calcium for activity. There are two major calpain isozymes: μ-calpain and m-calpain, which require micromolar or millimolar calicum for activity, respectively, in invitro assays. The major calpain isozymes are composed of two subunits: an 80kDa catalytic subunit; and a 30kDa regulatory subunit. The large and small subunits remain associated in the presence of calcium after proteolytic activation of the zymogen.

      Key Applications
      • ELISA
      • Western Blotting
      • Immunocytochemistry
      • Immunohistochemistry
Application NotesImmunoblot: 1:500-1,000. The small subunit of either μ-calpain m-calpain is 30kDa. ELISA: 1:50,000 (against the immunogen). Immunohistochemistry: 1:100 on 4% paraformaldehyde or 1% paraformaldehyde/4% glutaraldehyde fixed tissue. Immunocytochemistry: 1:100. Optimal working dilutions must be determined by the end user.

      Entrez Gene Number
Entrez Gene SummaryCalpains are a ubiquitous, well-conserved family of calcium-dependent, cysteine proteases. Calpain families have been implicated in neurodegenerative processes, as their activation can be triggered by calcium influx and oxidative stress. Calpain I and II are heterodimeric with distinct large subunits associated with common small subunits, all of which are encoded by different genes. This gene encodes a small subunit common to both calpain I and II and is associated with myotonic dystrophy. Two transcript variants encoding the same protein have been identified for this gene.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P04632 # Regulatory subunit of the calcium-regulated non- lysosomal thiol-protease which catalyzes limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction.
      COFACTOR: Calcium (By similarity).
SIZE: 268 amino acids; 28316 Da
SUBUNIT: Homodimer or heterodimer of a large (catalytic) and a small (regulatory) subunit. In presence of calcium, the heterodimer dissociates (By similarity).
SUBCELLULAR LOCATION: Cytoplasm (By similarity). Cell membrane (By similarity). Note=Translocates to the plasma membrane upon calcium binding (By similarity).
DOMAIN: SwissProt: P04632 The contact of the 5th EF-hand domain from each monomer allows the formation of the homodimer and also appears to mediate the contact between the large catalytic subunit and small regulatory subunit for the formation of the heterodimer (By similarity).
      SIMILARITY: Contains 5 EF-hand domains.
MISCELLANEOUS: EF-hand domains are paired. EF-hand 1 is paired with EF-hand 2 and EF-hand 3 is paired with EF-hand 4. The fifth EF-hand domain, left unpaired, does not bind the calcium but is responsible of the dimerization by EF-embrace. The first four EF- hand domains bind calcium, however it is not sure if the binding of EF-hand 4 to calcium is physiologically relevant.
