Background InformationCD99 antigen (UniProt P14209; also known as 12E7, E2 antigen, HBA-71 antigen, Protein MIC2, T-cell surface glycoprotein E2, Surface antigen MIC2) is encoded by the CD99 (also known as HBA71, MIC2, MIC2X, MIC2Y, MSK5X) gene (Gene ID 4267) in human. Originally identified as a thymus-leukemia-associated antigen, CD99 is a transmembrane glycoprotein expressed on many hematopoietic and non-hematopoietic cell types. CD99 is involved in cellular adhesion and migration as well as in survival. Antibody-mediated cross-linking of cell surface CD99 is shown to induce apoptotic signals in Jurkat T cells and essential thrombocythemia (ET) cell lines in vitro, and several CD99 monoclonal antibodies show in vivo efficacy in ET mouse models. CD99 is initially produced with a signal peptide sequence (a.a. 1-22), the removal of which yields the mature CD99 with a large extracellular domain (a.a. 23-122), followed by a transmembrane segment (a.a. 123-147) and a short cytoplasmic tail (a.a. 148-185).

      Applications
      Key Applications
      • Western Blotting
      • Flow Cytometry
      • Activity Assay
Application NotesWestern Blotting Analysis: 0.5 µg/mL from a representative lot detected CD99 in 10 µg of human thymus tissue lysate.
Flow Cytometry Analysis: 1.0 µg of this antibody from a representative lot detected CD99 in 1X10E6 THP-1 cells.
Activity Assay: A representative lot strongly diminished the recovery of undifferentiated normal B cell precursor (BCP) cells (BCP1), while being less effective against more differentiated BCP2 cells and exhibiting little effect toward mature BCP3 cells (Husak, Z., et al. (2010). J. Leukoc. Biol. 88(2):405-412).
Activity Assay: A representative lot affects the survival of TEL/AML1-positive acute lymphoblastic leukemia (ALL) cells in cultures, but not hyperdiploid or blasts with random karyotype (Husak, Z., et al. (2010). J. Leukoc. Biol. 88(2):405-412).
Activity Assay: A representative lot induced apoptosis (24 hrs) of mobilized peripheral blood (mPB) CD34+ cells in serum-free medium supplemented with IL-3, IL-6, and SCF, while no apoptosis induction by the mAb was detected (5 hrs) in the absence of cytokines (Imbert, A.M., et al. (2006). Blood. 108(8):2578-2586).
Activity Assay: A representative lot inhibited the transmigration of human monocytes through human bone marrow endothelial cell (HBMEC) monolayers in response to MCP-1, but not the transmigration of mobilized peripheral blood (mPB) CD34+ cells in response to SDF-1α or SDF-1α plus SCF (Imbert, A.M., et al. (2006). Blood. 108(8):2578-2586).
Activity Assay: A representative lot induced Mg++-dependent homotypic aggregation of Jurkat and CEMT, but not KE37, human T-cell lines, as well as CD4+/CD8+ human PBMCs, but not CD4+/CD8- or CD4-/CD8+ PBMCs, nor other non-T-cell lines tested (Bernard, G., et al. (1995). J. Immunol. 154(1):26-32).

SpecificityClone 0662 detects both the 32 kDa glycosylated (plasma membrane) and the 28 kDa non-glycosylated (intracellular) forms of human CD99/E2 (Gelin, C., et al. (1989). EMBO J. 8(11):3253-3259). Note that the clone was originally designated as 0662 (zero six six two) by Bernard, A., et al. (1988), but subsequently cited in some publications as O662 with a captial letter O.
