      DescriptionAnti-Atg4D Antibody
      Alternate Names
      • Cysteine protease ATG4D
      • AUT-like 4 cysteine endopeptidase
      • Autophagin-4
      • Autophagy-related cysteine endopeptidase 4
      • Autophagy-related protein 4 homolog D
Background InformationAtg4D (autophagy-related protein 4D) is a member of the Atg4 family of C54 endopeptidases that are required for the synthesis of autophagosomes. Atg4D is found to be cleaved by caspase-3 into a more active form where it can interact with the autophagosome marker Atg8. Atg4D is also known to be associated with mitochondrial dysfunction and the induction of apoptosis.

      Entrez Gene Number
Entrez Gene SummaryAutophagy is the process by which endogenous proteins and damaged organelles are destroyed intracellularly. Autophagy is postulated to be essential for cell homeostasis and cell remodeling during differentiation, metamorphosis, non-apoptotic cell death, and aging. Reduced levels of autophagy have been described in some malignant tumors, and a role for autophagy in controlling the unregulated cell growth linked to cancer has been proposed. This gene encodes a member of the autophagin protein family. The encoded protein is also designated as a member of the C-54 family of cysteine proteases. [provided by RefSeq].

      UniProt Number
UniProt SummaryFUNCTION: Cysteine protease required for autophagy, which cleaves the C-terminal part of either MAP1LC3, GABARAPL2 or GABARAP, allowing the liberation of form I. A subpopulation of form I is subsequently converted to a smaller form (form II). Form II, with a revealed C-terminal glycine, is considered to be the phosphatidylethanolamine (PE)-conjugated form, and has the capacity for the binding to autophagosomes.

      ENZYME REGULATION: Inhibited by N-ethylmaleimide.

      SUBCELLULAR LOCATION: Cytoplasm Probable.

TISSUE SPECIFICITY: Mainly expressed in skeletal muscle and, to a lower extent, in testis.

SEQUENCE SIMILARITIES: Belongs to the peptidase C54 family.
      Molecular Weight~56 kDa observed.
The calculated molecular weight is 53 kDa; however, Atg4D has been shown as a ~55 kDa band in western blots (Li, M., et al. (2011). J Biol Chem. 286(9):7327-7338.)
